Abstract ABSTRACT Objective:．To investigate the expression level of CD47 protein in laryngeal squamouscellcarcinomaanditsimpactonthedevelopmentofthecarcinomaandits clinical signifance,and explore the effect of silencing CD47 expression and anti-CD47 antibodies(anti-CD47mAbs) on the biological behaviors of laryngeal carcinomacellsHep-2. Method: 1.Thespecimenswere dividedintolaryngealsquamouscellcarcinoma group and normal laryngeal tissues group(control group).Western blot were used to detect the expression of CD47 protein.The relationship between CD47 expression level and its clinicapathological parameters was analyzed by SPSS 17.0. 2.CD47-RNAi-LV（CD47-RNAi-lentivirus）wastransfectedintoHep-2cells.Western blot was used to detect the expression of CD47protein of the Hep-2 cells pre- and post-RNAi,and chose the most effective RNAi. 3.Compare the influence of CD47-RNAi-LV and the anti-CD47mAbs on the cell proliferation ,apoptosis and invasionabilitybyM 、FCMandTranswellinvasionassayrespectively. Result:TheCD47proteinexpressioninlaryngealsquamouscellcarcinomaand itsnormal counterpartswere statisticallydifferent(p0.05).TheCD47expressionhad relationshipwiththe clinical stageand lymphnodemetastasiswhile notthe age and the degree of differentiation .Expression of CD47 protein was downregulated comparedwiththenegativegroupbyCD47RNAi.Comparedtonormalcontrolgroup, negativecontrolgroup,M showedthatfromthefourthdayontheRNAigroupand anti-CD47mAbsgrouphavethe similarRI(Rateofinhibition)(p0.05),therewasno statisticalsignificancebetweentheRNAigroupandanti-CD47mAbsgroup.Thecell apoptosisassayshowedthattheapoptosisrateoftheexprementalgroupswerehigher thanthe controlgroups.TheTranswellinvasionassayresults showedthatwithRNAi and anti-CD47 mAbs the number of Hep-2 cell penetrating significantly reduced,lowerthanthecontrolgroups.
