BackgroundandObjectives.Toexploretheroleofcis-regulatorysequenceswithintheβglobingeneclusteratchromosome11on humanγglobingeneexpressionrelatedtoHbEallele,weanalyzebaselinehematologicaldataandHbFvaluestogetherwithβ globinhaplotypesinhomozygousHbE.PatientsandMethods.80individualswithmolecularlycon?rmedhomozygousHbEwere analyzedfortheβglobinhaplotypesandXmnIpolymorphismusingPCR-RFLPs.74individualswithcompletelaboratorydata werefurtherstudiedforassociationanalyses.Results.Eightdi?erentβglobinhaplotypeswerefoundlinkedtoHbEalleles;three majorhaplotypeswere(a)(III),(b)(V),and(c)(IV)accountingfor94%ofHbEchromosomes.Anewhaplotype(Th-1)was identi?edandmostlikelyconvertedfromthemajorones.ThemajorityofindividualshadHbF5%;only10.8%ofhomozygous HbEhadhighHbF(average10.5%,range5.8–14.3%).Noassociationwasfoundonaspeci?chapl
