Flavivirusinfectionsarethemostprevalentarthropod-borneinfectionsworldwide,oftencausingseverediseaseespeciallyamong children,theelderly,andtheimmunocompromised.Intheabsenceofe?ectiveantiviraltreatment,preventionthroughvaccination wouldgreatlyreducemorbidityandmortalityassociatedwith?avivirusinfections.Despitethesuccessoftheempiricallydeveloped vaccinesagainstyellowfevervirus,Japaneseencephalitisvirusandtick-borneencephalitisvirus,thereisanincreasingneedfor amorerationaldesignanddevelopmentofsafeande?ectivevaccines.Severalbioinformatictoolsareavailabletosupportsuch rationalvaccinedesign.Indoingso,severalparametershavetobetakenintoaccount,suchassafetyforthetargetpopulation, overallimmunogenicityofthecandidatevaccine,ande?cacyandlongevity oftheimmuneresponsestriggered.Examplesof howbio-informaticsisappliedtoassistintherationaldesignandimprovementsofvaccines,particularly?avivirusvaccines,are presentedanddiscussed. 1.Introduction and antigenic di?erences.
