We investigatedwhetherhemodia?ltration(HDF)couldin?uencetheabilityofpolymorphonucleargranulocytes(PMGs)torelease NGAL.Theinvolvementofinterleukin-(IL-)1βandtumornecrosisfactor-(TNF-)αonNGALreleasewasevaluated.Methods. Westudiedend-stagerenaldisease(ESRD)patientsatthestartofdialysis(Pre-HDF)andattheendoftreatment(Post-HDF) and18healthysubjects(HSs).PeripheralvenousbloodwastakenfromHDFpatientsatthestartofdialysisandattheendof treatment.Results.PMGsobtainedfromESRDpatientswerehyporesponsivetoLPStreatment,withrespecttoPMGfromHS.IL- 1βandTNF-αproducedbyPMGfrompost-HDFpatientswerehigherthanthoseobtainedbyPMGfrompre-HDF.Neutralization ofIL-1β,butnotofTNF-α,determinedaclear-cutproductionofNGALinPMGfromhealthydonors.Onthecontrary,speci?c inductionofNGALinPMGfromuremicpatientswasdependentonthepresenceinsupernatantsofIL-1βandTNF-α.Conclusion.
