Nutrition Controls Mitochondrial Biogenesis in the Drosophila Adipose Tissue through Delg and Cyclin D/Cdk4 ClaudiaBaltzer1,2,StefanieK.Tiefenbo¨ck1,2,MarkMarti1¤,ChristianFrei1* 1DepartmentofBiology,ETHZurich,Zurich,Switzerland,2PhDprograminMolecularLifeSciences,Zurich,Switzerland Abstract Mitochondriaarecellularorganellesthatperformcriticalmetabolicfunctions:theygenerateenergyfromnutrientsbutalso providemetabolitesfordenovosynthesisoffattyacidsandseveralaminoacids.Thusmitochondrialmassandactivitymust becoordinatedwithnutrientavailability,yetthisremainspoorlyunderstood.Here,wedemonstratethatDrosophilalarvae growninlowyeastfoodhavestrongdefectsinmitochondrialabundanceandrespirationactivityinthelarvalfatbody.This correlates with reduced expression of genes encoding mitochondrial proteins, particularly genes involved in oxidative phosphorylation. Second, genes involved in glutamine metabolism are also expressed in a nutrient-dependent manner, suggesting a coordination of amino acid synthesis with mitochondrial abundance and activity. Moreover, we show that Delg(CG6338),theDrosophilahomologuetothealphasubunitofmammaliantranscriptionfactorNRF-2/GABP,isrequired forproperexpressionofmostgenesencodingmitochondrialproteins.OurdatademonstratethatDelgiscriticaltoadjust mitochondrialabundanceinrespecttoCyclinD/Cdk4,agrowth-promotingcomplexandglutaminemetabolismaccording tonutrientavailability.However,incontrasttonutrients,Delgisnotinvolvedintheregulationofmitochondrialactivityin thefatbody.Thesefindingsarethefirstgeneticevidencethattheregulationofmitochondrialmasscanbeuncoupledfrom mitochondrialactivity.
