Results:Wecombinedhighaccuracymassspectrometryandbioinformaticapproachestopresentforthefirsttime anexperimentallydeterminedcorenuclearproteomeforP.falciparum.Besidesalargenumberoffactors implicatedinknownnuclearprocesses,one-thirdofalldetectedproteinscarrynofunctionalannotation,including manyphylum-orgenus-specificfactors.Importantly,extensiveexperimentalvalidationusing30transgeniccell linesconfirmedthehighspecificityofthisinventory,andrevealeddistinctnuclearlocalizationpatternsofhitherto uncharacterizedproteins.Further,ourdetailedanalysisidentifiednovelproteindomainspotentiallyimplicatedin genetranscriptionpathways,andshedsimportantnewlightonnuclearcompartmentsandprocessesincluding regulatorycomplexes,thenucleolus,nuclearpores,andnuclearimportpathways. Conclusion:OurstudyprovidescomprehensivenewinsightintothebiologyofthePlasmodiumnucleusandwill serveasanimportantplatformfordissectinggeneralandparasite-specificnuclearprocessesinmalariaparasites.
