However,themolecularmechanismofPLPfortheinhibitionofliverfibrosisisunclear.Thisstudyaimsto investigatethemolecularproteinsignaturesinvolvedinthehepatoprotectivemechanismsofPLPviaaproteomics approachusingathioacetamide(TAA)-inducedliverfibrosisratmodel. Methods:MaleSprague–Dawleyratsweredividedintothreegroupsofsixasfollows:Normalgroup;TAAgroup,in whichratsreceivedTAAonly;andPLPgroup,inwhichratsreceivedPLPandTAA.Liverfibrosiswasinducedinthe ratsbyrepeatedintraperitonealinjectionsofTAAatadoseof200mg/kgbodyweighttwiceaweekfor4weeks. PLPwasgivenorallyatadoseof50mg/kgbodyweighttwiceadayfromthebeginningoftheTAAtreatment untiltheendoftheexperiment.Thedevelopmentoflivercirrhosiswasverifiedbyhistologicalexamination.Liver proteomeswereestablishedbytwo-dimensionalgelelectrophoresis.Proteinswithsignificantlyalteredexpression levelswereidentifiedbymatrix-assistedlaserdesorption/ionization-timeofflight/timeofflightmassspectrometry andthedifferentiallyexpressedproteinswerevalidatedbyimmunohistochemicalstainingandreversetranscription polymerasechainreaction. Results:HistologicalstainingshowedaremarkablereductioninliverfibrosisintheratswithPLPtreatment.Atotal of13differentiallyexpressedproteinsincludingactin,tubulinalpha-1Cchain,preprohaptoglobin,hemopexin, galectin-5,glutathioneS-transferasealpha-4(GSTA4),branchedchainketoaciddehydrogenasehterotetramericE1 subunitalpha(BCKDHA),glutathioneS-transferasemu(GSTmu);glyceraldehyde-3-phosphatedehydrogenase (GAPDH);thiosulfatesulfurtransferase(TFT);betaine-homocysteineS-methyltransferase1(BHMT1);quinoid dihydropteridinereductase(QDPR
