Mirbolookietal.EJNMMIResearch2011,1:30 /content/1/1/30 ORIGINAL RESEARCH OpenAccess Quantitativeassessmentofbrownadiposetissue 18 metabolicactivityandvolumeusing F-FDG PET/CTandβ3-adrenergicreceptoractivation MRezaMirbolooki,CristianCConstantinescu,Min-LiangPanandJogeshwarMukherjee* Abstract Background:Brownadiposetissue[BAT]metabolisminvivoisvitalforthedevelopmentofnovelstrategiesin combatingobesityanddiabetes.Currently,BATisactivatedatlowtemperaturesandmeasuredusing2-deoxy-2- 18F-fluoro-D-glucose[18F-FDG]positron-emissiontomography[PET].Wereporttheuseofb3-adrenergicreceptor- mediatedactivationofBATatambienttemperaturesusing(R,R)-5-[2-[2,3-(3-chlorphenyl)-2-hydroxyethyl-amino] propyl]-1,3-benzodioxole-2,2-dicarboxylate,disodiumsalt[CL316,243](aselectiveb3-adrenoceptoragonist)and measuredby 18F-FDGPET/computedtomography[CT]. Methods:ControlandCL316,243-treated(2mg/kg)maleSprague-Dawleyratswereadministeredwith 18F-FDGfor PET/CTstudiesandwerecomparedtoanimalsatcoldtemperatures.Receptor-blockingexperimentswerecarried outusingpropranolol(5mg/kg).DoseeffectsofCL316,243werestudiedbyinjecting0.1to1mg/kg30minprior to 18F-FDGadministration.Imagingresultswereconfirmedbyautoradiography,andhistologywasdonetoconfirm BATactivation. Results:CL316,243-activatedinterscapularBAT[IBAT],cervical,periaortic,andintercostalBATswereclearly visualizedbyPET. 18F-FDGuptakeofIBATwasincreased12-foldbyCL316,243vs.1.1-foldbycoldexposurewhen comparedtocontrols. 18F-FDGuptakeoftheCL-activatedIBATwasreducedby96.0%usingintraperitoneal administrationofpropranolol.Average 18F-FDGuptakeofIBATincreased3.6-,3.5-,and7.6-foldbydosesof0.1,0.5, and1mg/kgCL,respectively.Exvivo 18F-FDGautoradiographyandhistologyoftransversesectionsofIBAT confirmedintenseuptakeintheCL-activatedgroupandactivatedIBATvisualizedbyPET. Conclusion:OurstudyindicatedthatBATmetabolicactivitycouldbeevaluatedby 18F-FDGPETusingCL316,243 atambienttemperatureintherodentmodel.Thisprovidesafeasibleandreliableme
