Re-EvaluatetheEffectofHyperbaricOxygenTherapyin Cancer-APreclinicalTherapeuticSmallAnimalModel Study SnehaPande1,AmitSengupta1*,AnuragSrivastava2,RajivP.Gude3,ArvindIngle4 1BioengineeringLaboratory,AdvancedCentreforTreatment,ResearchEducationinCancer,TATAMemorialCentre,NewMumbai,Maharashtra,India,2Departmentof Cancer Surgery, All India Institute ofMedical Science, New Delhi, India, 3Gude Laboratory, Advanced Centre for Treatment, Research and Education in Cancer,TATA MemorialCentre,NewMumbai,Maharashtra,India,4LaboratoryAnimalFacilities,AdvancedCentreforTreatment,ResearchandEducationinCancer,TATAMemorial Centre,NewMumbai,Maharashtra,India Abstract Tumorhypoxiaisaknowndriverofangiogenesisthatalsofacilitatestumorgrowth.Moreover,poorlyoxygenatedcentral tumorarearemainsrelativelyradioorchemoresistant.HBOtherapyisknowntoelevatethelevelsofdissolvedoxygenand eliminatestumorhypoxia.Ithasbeenoneofthemodalitiesincancertreatment;thereforeitsoptimizationisimportant.In thisexperimentalstudy,nocancerenhancingeffectwasseenduringthecourseofHBOtherapy;however,posttherapy therewasanacceleratedgrowthandprogressionoftumor.HBOtreatedmicelivedshorterandtheresponsetotherapywas dosetumorvolumedependent.HBOtherapyprobablyexertitseffectonthecancerproliferatingcellsthroughmultiple pathwayssuchasincreasedDNAdamage,apoptosisgeno-toxicityleadingtoslowcancerprogressionwhileposttherapy tumorigeniceffectcouldbeduetoimpairedDNArepairmechanism,mutageniceffectaneuploidyaswellasalteredblood supplynutrients.Tumorgrowthreachedplateauwithtimeandthisfindingvalidatedtheoreticalmodelpredictingtumor reachinganasymptoticlimit.While,markedasymmetryobservedintumorvolumeprogressionorcancercellproliferation rateineachoftheexperimentalC3Hmousesuggestedaneedforanalternatesmallanimalpre-clinicalcancertherapeutic model.
