Methods and Findings: Eighty white rabbits were randomly divided into four groups. One eye from each rabbit was randomlychosentoreceivetopical0.1%BACtwicedailyfor2weeks(GroupBAC-W2),3weeks(GroupBAC-W3),4weeks (GroupBAC-W4),or5weeks(GroupBAC-W5).Fluoresceinstaining,Schirmer’stests,andconjunctivalimpressioncytology wereperformedbeforeBACtreatment(normal)andondays0,7,14and21afterBACremoval.Theeyeballswerecollected atthesetimepointsforimmunofluorescencestaining,hematoxylinandeosin(HE)staining,andelectronmicroscopy.After removingBAC,thesignsofdryeyeinGroupBAC-W2lastedoneweek.Comparedwithnormal,therewerestillsignificant differencesintheresultsofSchirmer’stestsandfluoresceinstaininginGroupsBAC-W3andBAC-W4onday7(P,0.05 )and inGroupBAC-W5onday14(P,0.05).Decreasesingobletcelldensityremainedstableinthethreeexperimentalgroupsat all time points (P,0.001). Decreased levels of mucin-5 subtype AC (MUC5AC), along with histopathological and ultrastructuraldisordersofthecorneaandconjunctivacouldbeobservedinGroupBAC-W4andparticularlyinGroupBAC- W5untilday21. Conclusions:Astablerabbitdryeyemodelwasinducedbytopical0.1%BACfor5weeks,andafterBACremoval,thesigns ofdryeye
