rstTranscriptionalActivityInfluenceskirremRNA ConcentrationintheDrosophilaPupalRetinaduringthe FinalStepsofOmmatidialPatterning MaiaroCabralRosaMachado,ShirleiOctacilio-Silva¤,MaraSilviaA.Costa,RicardoGuelermanP.Ramos* DepartmentodeBiologiaCelulareMoleculareBioagentesPatoge?nicos,FaculdadedeMedicinadeRibeira?o Preto,UniversidadedeSa?o Paulo,Ribeira?o Preto,Sa?o Paulo, Brazil Abstract Background:Drosophilaretinalarchitectureislaiddownbetween24–48hoursafterpupariumformation,whensomeof thestilluncommittedinterommatidialcells(IOCs)arerecruitedtobecomesecondaryandtertiarypigmentcellswhilethe remainingonesundergoapoptosis.Thischoicebetweensurvivalanddeathrequirestheproductoftheroughest(rst )gene, animmunoglobulinsuperfamilytransmembraneglycoproteininvolvedinawiderangeofdevelopmentalprocesses.Both temporalmisexpressionofRstandtruncationoftheproteinintracytoplasmicdomain,leadtoseveredefectsinwhichIOCs either remain mostly undifferentiated and die late and erratically or, instead, differentiate into extra pigment cells. Intriguingly,mutantsnotexpressingwildtypeproteinoftenhavenormalorverymildrougheyes. Methodology/PrincipalFindings:ByusingquantitativerealtimePCRtoexaminersttranscriptionaldynamicsinthepupal retina,bothinwildtypeandmutantallelesweshowedthattightlyregulatedtemporalchangesinrsttranscriptionalrate underlieitsproperfunctionduringthefinalstepsofeyepatterning.Furthermorewedemonstratedthattheunexpected wildtypeeyephenotypeofmutantswithlowornorstexpressioncorrelateswithanupregulationinthemRNAlevelsofthe rst paralogue kin-of-irre (kirre), which seems able to substitute for rst function in this process, similarly to their role in myoblastfusion.ThiscompensatoryupregulationofkirremRNAlevelscouldbedirectlyinducedinwildtypepupaupon RNAi-mediated silencing of rst, indicating that expression of both genes is also coordinately regulated in physiological conditions.
