Methods:1,016subjectsaged70yearswereinvestigatedinthePIVUSstudy.Fourphthalatemetaboliteswere detectedintheserumofalmostallsubjects(96%)byanAPI4000liquidchromatograph/tandemmass spectrometer.AbdominalMRIwasperformedinarepresentativesubsampleof287subjects(28%),andadual- energyX-rayabsorptiometry(DXA)-scanwasobtainedin890(88%)ofthesubjectstwoyearfollowingthe phthalatemeasurements. Results:Inwomen,circulatingconcentrationsofmono-isobutylphthalate(MiBP)werepositivelyrelatedtowaist circumference,totalfatmassandtrunkfatmassbyDXA,aswellastosubcutaneousadiposetissuebyMRI followingadjustmentforserumcholesterolandtriglycerides,education,smokingandexercisehabits(allp0.008). Mono-methylphthalate(MMP)concentrationswererelatedtotrunkfatmassandthetrunk/leg-ratiobyDXA,but lesspowerfulthanMiBP.However,nosuchstatisticallysignificantrelationshipswereseeninmen. Conclusions:ThepresentevaluationshowsthatespeciallythephthalatemetaboliteMiBPwasrelatedtoincreased fatamountinthesubcutaneousabdominalregioninwomenmeasuredbyDXAandMRItwoyearslater.
