Pantazakietal.AMBExpress2011,1:17 /content/1/1/17 ORIGINAL OpenAccess Simultaneouspolyhydroxyalkanoatesand rhamnolipidsproductionbyThermusthermophilus HB8 AnastasiaAPantazaki1*,ChristosPPapaneophytou1andDimitraALambropoulou2 Abstract TheabilityofThermusthermophilusHB8toproducesimultaneouslytwoenvironmentally-friendlybiodegradable products,polyhydroxyalkanoates(PHAs)andrhamnolipids(RLs),usingeithersodiumgluconateorglucoseassole carbonsource,wasdemonstrated.TheutilizationofsodiumgluconateresultedinhigherlevelsofPHAsandRLs productionthanwhenglucosewasusedassolecarbonsource.Theinitialphosphateconcentration(asPO43-) influencesbothPHAsandRLsproductionsthatwereincreasedduringcultivationtime.PHAsaccumulationwas enhanced(300mg/L)after72hofcultivationinaninitial[PO43-]of25mM,whileRLsproduction(200mg/L) wasstartedafter35handcontinueduntil72hofcultivation,inaphosphate-limitedmediumcontaininginitially5 mMof[PO43-].Inaddition,thecombineeffectofinitial[PO43-]andcultivationtimeonbiomass,PHAsandRLs productionwasevaluatedfrom2Dcontourplots.Theresultsrevealedthatlowinitialphosphateconcentrations (upto5mM)andlongincubationtime(72h)promotedRLsbiosynthesiswhilehigherinitialphosphate concentrations(upto25mM)wherefavorableforbiomassandPHAsproduction.Themolecularcompositionof theproducedbio-productswasidentified.TheaccumulatedPHAswereco-polymerswhichmainlyconsistedof3- hydroxydecanoate(3HD)asresultedbygaschromatography(GC)analysis.ThesecretedRLswereextractedand theirtotalmixturecontainedbothmono-anddi-RLsidentifiedbythin-layerchromatography(TLC).Moreover,the molecularcompositionoftheproducedRLscharacterizedindetailsbyLC-MSanalysisshowedaplethoraof diversityincludingmono-,anddi-RLs,di-rhamno-monolipidiccongenersdifferinginthelengthofthelipidicchain, whichadditionallywerefoundtobesaturatedorunsaturatedinsomecases.
