This is primarilybecausetherehasbeennoreliablemolecularmethodforestimatingorquantifyingvariation. Methodology/PrincipalFindings:Weadoptedapetal-variegatedcultivarofSaintpaulia‘Thamires’(Saintpauliasp.)asthe modelplant.Basedonthedifferencebetweenthepre-andpost-transposonexcisionsequenceofthepromoterregionof flavonoid39,59-hydoroxylase(F3959H),weestimatedmutated(transposon-excised)cellpercentagesusingaquantitativereal- timePCR.Mutatedcellpercentagesinleaflaminaeusedasexplantswas4.6and2.4%inhighlyorlowvariegationflower plants,respectively,althoughtheoccurrencesofbluecolormutantsintheirregenerantsweremorethan40%.Preexisting mutatedcell percentages inculturedexplants wereconsiderably lower thanthemutatedplant percentageamongtotal regenerantsviatissueculture. Conclusions/Significance: The estimation of mutated cell percentages became possible using the quantitative real-time PCR. The origins of mutations were successfully distinguished; it was confirmed that somaclonal variations are mainly causedbynewlygeneratedmutationsarisingfromtissuecultureprocess.
