This is accomplishedthroughtheproliferationandsubsequentdifferentiationoftheadultstemcells.Thisself-renewalsystemis established in the so-called postembryonic developmental period in mammals when endogenous thyroid hormone (T3) levelsarehigh. Methodology/Principal Findings: The T3-dependent metamorphosis in anurans like Xenopus laevis resembles the mammalianpostembryonicdevelopmentandoffersauniqueopportunitytostudyhowtheadultstemcellsaredeveloped. Thetadpoleintestineispredominantlyamonolayeroflarvalepithelialcells.Duringmetamorphosis,thelarvalepithelialcells undergoapoptosisand,concurrently,adultepithelialstem/progenitorcellsdevelopdenovo,rapidlyproliferate,andthen differentiatetoestablishatrough-crestaxisoftheepithelialfold,resemblingthecrypt-villusaxisintheadultmammalian intestine.Theleucine-richrepeat-containingGprotein-coupledreceptor5(LGR5)isawell-establishedstemcellmarkerin theadultmouseintestinalcrypt.HerewehaveclonedandanalyzedthespatiotemporalexpressionprofileofLGR5gene duringfrogmetamorphosis.WeshowthatthetwoduplicatedLGR5genesinXenopuslaevisandtheLGR5geneinXenopus tropicalisarehighlyhomologoustotheLGR5inothervertebrates.Theexpre
