AlthoughglobalawarenessregardingHIV-1/HCVco-infectionisincreasinglittleisknownaboutthepathophysiologythat mediatestherapidprogressiontohepaticdiseaseintheco-infectedindividuals. Results:Inthisstudy,weinvestigatedtheproteomeprofilesofperipheralbloodmononuclearcellsfromHIV-1mono-, HCVmono-,andHIV-1/HCVco-infectedpatients.Theresultsofhigh-resolution2DgelelectrophoresisandPDquest softwarequantitativeanalysisrevealedthatseveralproteinsweredifferentiallyexpressedinHIV-1,HCV,andHIV-1/HCVco- infection.Liquidchromatography-massspectrometryandMascotdatabasematching(LC-MS/MSanalysis)successfully identified29uniqueanddifferentiallyexpressedproteins.Theseincludedcytoskeletalproteins(tropomyosin,gelsolin, DYPLSL3,DYPLSL4andprofilin-1),chaperonesandco-chaperones(HSP90-betaandstress-inducedphosphoprotein), metabolicandpre-apoptoticproteins(guanosinetriphosphate[GTP]-bindingnuclearproteinRan,thedetoxifyingenzyme glutathioneS-transferase(GST)andRhoGDP-dissociationinhibitor(Rho-GDI),proteinsinvolvedincellprosurvival mechanism,andthoseinvolvedinmatrixsynthesis(collagenbindingprotein2[CBP2]).Thesixmostsignificantand relevantproteinswerefurthervalidatedinagroupofmono-andco-infectedpatients(n=20)atthetranscriptionallevels. Conclusions:Thespecificpro-andanti-apoptoticproteinsignaturesrevealedinthisstudycouldfacilitatethe understandingofapoptoticandprotectiveimmune-mediatedmechanismsunderlyingHIV-1andHCVco-infectionand theirimplicationsonliverdiseaseprogressioninco-infectedpatients.
