Soitamoetal.BMCPlantBiology2012,12:204 /1471-2229/12/204 RESEARCH ARTICLE OpenAccess ExpressionofgeminiviralAC2RNAsilencing suppressorchangessugarandjasmonate responsivegeneexpressionintransgenictobacco plants ArtoJSoitamo*,BalajiJadaandKirsiLehto Abstract Background:RNA-silencingisaconservedgeneregulationandsurveillancemachinery,whichinplants,isalso usedasmajordefencemechanismagainstviruses.Variousvirus-specificdsRNAstructuresarerecognizedbythe silencingmachineryleadingtodegradationoftheviralRNAsor,asincaseofbegomoviruses,tomethylationof theirDNAgenomes.VirusesproducespecificRNAsilencingsuppressor(RSS)proteinstopreventthesehostdefence mechanisms,andastheseinterferewiththesilencingmachinerytheyalsodisturbtheendogenoussilencing reactions.Inthispaper,wedescribehowexpressionofAC2RSS,derivedfromAfricancassavamosaicgeminivirus changestranscriptionprofileintobacco(Nicotianatabacum)leavesandinflowers. Results:ExpressionofAC2RSSintransgenictobaccoplantsinducedclearphenotypicchangesbothinleavesand inflowers.Transcriptomesoftheseplantswerestronglyaltered,withtotalof1118and251differentiallyexpressed genesinleavesandflowers,respectively.Thethreemostup-regulatedtranscriptgroupswererelatedtostress,cell wallmodificationsandsignalling,whereasthethreemostdown-regulatedgroupswererelatedtotranslation, photosynthesisandtranscription.Itappearsthatmanyofthegeneexpressionalterationsappearedtoberelatedto enhancedbiosynthesisofjasmonateandethylene,andconsequentenhancementofthegenesandpathwaysthat areregulatedbythesehormones,ortotheretrogradesignallingcausedbythereducedphotosyntheticactivity andsugarmetabolism.ComparisonoftheseresultstoaprevioustranscriptionalprofilingofHC-ProRSS-expressing plantsrevealedthatsomeofsamegeneswereinducedbybothRSSs,buttheirexpressionlevelsweretypically higherinAC2thaninHC-ProRSSexpressingplants.Allinall,alargenumberoftranscriptalterationswerefoundto bespecifictoeachoftheRSSexpressingtransgenicplants.
