组比较差异无显著性( P＞0.05 ) 。BGC-823 细胞72h 各药物浓度组的凋亡率与阴 性对照组比较及组间比较差异均有显著性( P 0.05 ) ，24h 、48h 各药物浓度组与 阴性对照组比较差异无显著性( P ＞0.05 ) ；（4 ）RT-PCR 检测人胃癌细胞株 SGC-7901、MKN-28 无RASSF1AmRNA 表达，经 5-Aza-CdR 处理后基因重新表达， BGC-823 处理前后RASSF1AmRNA 均有表达。 结论：1、RASSF1A 抑癌基因沉默与胃癌的发生相关；2、5-Aza-CdR 可以 抑制细胞 SGC-7901、MKN-28 细胞的增殖，改变细胞周期，促进细胞凋亡。3、 5-Aza-CdR 对BGC-823 细胞，明显抑制细胞增殖，并促进细胞凋亡，对生长周期 无显著影响。4、 5-Aza-CdR 可能系通过消除RASSF1A 基因抑癌基因的启动子 甲基化状态，使其重新表达而抑制胃癌细胞的生长；并促进凋亡。 关键词：5-氮杂2-脱氧胞苷 胃癌细胞株 RASSF1A 基因 3 Effect of 5-Aza-2-deoxycytidine on human gastric carcinoma cell lines with analysis of suppressive role of RASSF1A gene Abstract Objective: To observe the effects of specificity methyltransferases inhibitor 5-aza-2’ -deoxycitydine (5-Aza-CdR) on the proliferation of human gastric cancer cell lines BGC823、SGC7901 and MKN-28as well asd epression of RASSF1A gene. Methods: Human gastric cancer cell lines BGC823、SGC7901 and MKN-28 were cultured in RPMI1640 and then treated with different concentrations of 5-Aza-CdR (0.4，1.6， 6.4，25.6，102.4 μmol/L). The proliferation of the cells was detected by MTT assay and flow cytometry(FCM). the expression of RASSF1A gene in the three kinds of cell lines was detected by reverse transcription-polymerase chain reaction (RT-PCR). Result: 5-Aza-CdR displayed a growth inhibitory effect on BGC823、SGC7901 and MKN-28 cells in a dose- and time-dependent manner after exposure to 5-Aza-CdR at different concentrations(0.4，1.6，6.4，25.6，102.4 μmol/L)for 24,48 and 72 hours.
