48/80 刺激的MCs 的残余物组与DMEM ＋Compound 48/80 组相比，差异具有显 著性（p0.05 ）；Compound 48/80 刺激的MCs 的颗粒释放液组和Compound 48/80 刺激的MCs 的残余物组相比，差异具有显著性（p0.05 ）。 3 FITC 标记的鬼笔环肽观察VSMCs 微丝结构的改变，结果显示： (1) MCs 组与对照组相比，VS MCs 微丝纤细，稀疏，荧光强度降 低，应力纤维部分解聚。 (2) MCs 颗粒释放液组的 VS MCs 微丝明显变细，稀疏，密度降低， 荧光强度明显降低，细胞外周的应力纤维解聚，中央微丝束明显增加。 结论 (1) Compound 48/80 和富马酸酮替芬对 ApoE -/ - 小鼠血脂含量无 明显影响, Compound 48/80 促进 ApoE -/ - 小鼠的胸主动脉 AS 斑块的 形成，而富马酸酮替芬对 ApoE -/ - 小鼠的胸主动脉 AS 斑块具有一定抑 制作用。 (2 ) MC s 能促进 VS MCs 发生迁移,Compound 48/80 刺激的 MCs 能明显促进 VS MCs 发生迁移，其中 MCs 的颗粒释放液是 MCs 促进 VS MCs 发生迁移的主要原因。 (3) MCs 颗粒释放液促进 VS MCs 细胞骨架微丝重排，VS MCs 迁 移能力增加。 关键词： Compound 48/80 肥大细胞 血管平滑肌细胞 ApoE 基因敲除 小鼠 动脉粥样硬化 3 Effects of Compound 48/80 on the plaque in the ApoE-/- mice aorta and the study of mast cell on the migration of smooth muscle cell ABSTRACT Objective: Mast cells （MCs ）are a kind of inflammatory-immune cells. Recently, with the progress of atherosclerosis(AS) etiology, we found that MCs were involved in the every evolutive phase of atherosclerosis. Vascular smooth muscle cells (VSMCs), migrating from the media to the intima, and some inflammatory-immune cells(T lymphocyte, macrophage, mast cells and so on) are the cellar components of as lesions. But the relationship between MCs and VSMCs has not been clarified.
