Effect of Ca b Subunits on Structural Organization of v Cav 1.2 Calcium Channels . . Evgeny Kobrinsky , Parwiz Abrahimi , Son Q. Duong, Sam Thomas, Jo Beth Harry, Chirag Patel, Qi Zong Lao, Nikolai M. Soldatov* National Institute on Aging, National Institutes of Health, Baltimore, Maryland, United States of America Abstract Background: Voltage-gated Ca 1.2 calcium channels play a crucial role in Ca2+ signaling. The pore-forming a subunit is v 1C regulated by accessory Ca b subunits, cytoplasmic proteins of various size encoded by four different genes (Ca b - b ) and v v 1 4 expressed in a tissue-specific manner. Methods and Results: Here we investigated the effect of three major Ca b types, b , b and b , on the structure of Ca 1.2 v 1b 2d 3 v in the plasma membrane of live cells. Total internal reflection fluorescence microscopy showed that the tendency of Cav 1.2 to form clusters depends on the type of the Ca b subunit present. The highest density of Ca 1.2 clusters in the plasma v v membrane and the smallest cluster size were observed with neuronal/cardiac b present. Ca 1.2 channels containing b ,
