The Expression of ABCA1 in Monocytes of Patients with
Coronary Heart Disease and Effects of Fenobrate
on the Expression of ABCA1
ABSTRACT
Objective To investigate the relationship between the degree of cornary
atherosclerosis and the expression of ATP binding cassette transporter
A1(ABCA1) and the effects of fenobrate on the expression of ABCA1 in
monocytes of coronary heart disease(CHD) patients.
Methods People who underwent coronary angiography were divided
into different groups according to the severity of cornary sterosis.Serum
lipids were measured in all subjects.Monocytes were isolated by density
gradient method from blood of samples.Total RNA of monocytes were
abstracted and reverse transcription polymerase chain reaction (RT-PCR)
were performed to inspect the expression of ABCA1 mRNA in different
groups.Western blot were performed to inspect the expression of ABCA1
protein level in different groups.Monocytes in CHD groups were incubated
with fenobrate. The mRNA and protein level of PPARα and ABCA1 were
inspected.
Results There were stastistic difference at the level of TC、LDL-c、
HDL-c in control group and CHD groups(P0.05). There were no stastistic

difference at the expression of ABCA1 mRNA and protein in control group
and CHD groups(P0.05). Fenobrate can increase mRNA level of PPARα
and ABCA1 in monocytes of CHD patients (P0.05).Fenobrate can
upgraduate the protein expression of PPARα and ABCA1 in monocytes of
CHD patients (P0.05).
Conclusions The increased level of HDL-c and the decreased level of
TC and LDL-c were correlated with the s
