为87μg/ml~550μg/ml，平均值为（206.45±179.39）μg/ml（n=5）。8.人外周血中ARMET的水平成年男女正常血清中未检测到ARMET（男20名，女20名），100例肿瘤患者血清中未检测到ARMET。结论已成功制备特异性较高的抗人ARMET单克隆抗体及ELISA检测试剂盒，后者可测范围广，灵敏度和精密度均较高。关键词：ARMET/MANF/ARP；单克隆抗体；杂交瘤；ELISA检测方法英文摘要PreparationofmonoclonalantibodiesagainsthumanARMETandestablishmentofdetectionmethodbyELISAPH.Mcandidate:WangFacaiSupervisor:Prof.ShenYuxian（TheKeyLaboratoryofGeneResourceUtilizationforGenneticMinistryofEducationandAnHuiProvience,InstituteofClinicalPharmacology,AnhuiMedicalUniversity,Hefei230032)ABSTRACTInpublicdatabase,ARMET(Argininerich,mutatedinearlystageoftumors)wasalsodesignatedasARP(arginine-richprotein)orMANF(mesencephalicastrocyte-derivedneurotrophicfactor).Sofar,onlyfewreportsassociatedwithARMETareavailabeanditsdetailedfunctionisstillunclear.Amongthe30000geneswescreenedbymicroarray,ARMETwasthemostsensitiveoneupregulatedbyendoplasmicreticulumstress(ER-stress).OurrecentdataindicatesthatARMET,inducedbycerebralischemia,iswidelyexpressedinneuronsandgliacells.However,notlikeitsdenomination,astrocytesarenotthemainsourceofARMET.Therefore,wethinkMANFisnotapropernameforARMET.SinceERstressisinvolvedintheon-setofmanydiseases,ARMET,asasecretedprotein,maybeanserologicalmarkerfortheearlydiagnosis.Therefore,itisveryimportanttoestablishagoodrapidclinicserologicaldetectionmethodforARMET.OBJECTIVE:TopreparemonoclonalantibodiesagainsthumanARMET(mAb)andestablishstandardprocedureforARMETdetectionbyELISA.METHODS:pET28a-ARMETwastransformedintoE.coliBL21.TheHis-taggedfusionproteinwasexpressedunderIPTGinductionandfurtherprocessedforaffinitypurificationbynicklebeads.AfterrunningSDSgelandstainingCommassieBlue,theimagesweretakenandthepurityofARMETwasanalyzedbyBandScan.TheconcentrationofARMETwasmeasuredbybicinchoninicacidproteinassays(BCA).FemaleBALB/cmicewereimmunizedwithpurifiedARMETprotein.ThemAbagainstARMETwaspreparedbyhybridomatechnology.ThehybridomacellsproducingmAbsagainstARMETwereinjectedintomouseperitonealcavityandmAbswereharvestedfromasciticfluid.IndirectELISAwasusedtoc
