关键词：免疫球蛋白M；血管生成；喉肿瘤；癌；VEGF；CD40EffectandmechanismofAnti-humanIgMantibodyontheangiogenesisofhumanlaryngealsquamouscellcarcinomaHep-2cellinnudemiceAbstract:Objective:LaryngealcarcinomaisamalignancythatoriginatefromepithelialtissueofLaryngealmucousmembranein.About5.7%-7.6%insystemictumors.Theincidenceratehasgraduallyincreasedinrecentyears,nowthemaintreatmentmodeofLaryngealcarcinomaaresurgery,radiotherapyandchemotherapy.butthetreatmenteffectisstillnotsatisfied.The5yearsurvivalrateisnotenhanceddramatically.withthedevelopmentofbiologytechnology,peopleareseekingnewtreatmentmodeandnewdrugtotreatlaryngealcarcinoma.Recentyearsmanyresearchersdiscoveredsomeepitheliogenicmalignanttumorcanexpressimmunoglobulin(Ig).Thisphenomenonhasbecomeahotspotinimmunestudyoftumor,andmayprovideanewthoughtfortumor’streatment.Tumorangiogenesiscorrelatedwithtumor‘soccurrenceanddevelopment.Theexpressionofangiogensisfactoristhemostimportantstepintumor’sangiogensis.Inthisstudyweinvestigatedtheeffectofanti-humanIgMantibodyontheangiogenesisoflaryngealsquamouscellcarcinomaHep-2celltransplantedtumorestablishedinnudemice,VEGF,CD40proteinexpressionandthemicrovesseldensity(MVD)ofthetumor.soastoexploretheanti-angiogenesiseffectofanti-humanIgMantibodyandthepossiblemechanism,soastoprovidetheoreticalandexperimentalevidencefortheuseofanti-humanIgMantibodyinthetreatmentoflaryngealsquamouscellcarcinoma.Methods:Inourearlyresearchwehadestablishedthetransplantedmodeofhumanlaryngealsquamouscellcarcinomainnudemice.Thenwedividedthe15miceinto3groupsrandomly:experimentalgroup(IgMgroup),controlgroupofIgGandcontrolgroupofPBS.Inexperimentalgroupweinjectedanti-humanIgMantibodyintransplanttumortissue.everytimeeachnudemousewasinjected0.3mg,inevery3days,thereare5timesintotal.controlgroupofIgG(treatedwithnormalgoatIgG),.controlgroupofPBS(treatedwithnormalPBS),themethodofinjectionissameasexperimentalgroup.Weobservethenudemice’sgrowthconditionthenwekilledthemiceatday3afterlasttreatment,andgettheTumortissue.Weuseimmunohistochemicalte
