摘 要 目的：通过检测变应性鼻炎(Allergic rhinitis AR)患者外周血白细胞介素 35 （IL-35）及其亚基 EBI3 mRNA、IL-12A mRNA 的表达探讨其与 AR 的发生的 关系及临床意义。 方法：分别采集变应性鼻炎患者（46 例）及正常对照组（30 例）外周血， 采用酶联免疫吸附（ELISA）法检测血清 IL-35 表达水平，同时用 SYBR Green 实时定量 PCR（qPCR）检测外周血单个核细胞中 IL-35 亚基 EBI3 mRNA 及 IL-12A mRNA 的表达水平。 结果：AR 患者组血清中 IL-35 的水平为 251.22±46.27pg/ml,正常对照组血清 中 IL-35 水平为 382.17±25.41pg/ml，AR 组血清 IL-35 水平明显低于正常对照组， 差异具有统计学意义（p0.01）。外周血单个核细胞 EBI3 mRNA 的表达水平 AR 组明显低于正常对照组,AR 组 EBI3 mRNA 的表达量约为正常对照组的一半，差 异具有统计学意义（p0.001）。而外周血单个核细胞 IL-12A mRNA 的表达水平 AR 组与正常对照组无统计学差异(p0.05)。 结论：AR 患者体内 IL-35、EBI3 mRNA 水平降低可能与疾病的发生和发展 密切相关。 关键词：变应性鼻炎；白细胞介素 35；EBI3 mRNA；IL-12A mRNA II  ABSTRACT Objective：To explore the expression of interleukin-35(IL-35)、Epstein-Barr virus-induced gene 3(EBI3) mRNA and IL-12A mRNA in peripheral blood of patients with allergic rhinitis and its significance. Methods：Peripheral blood Were collected from patients with allergic rhinitis (46 cases) and healthy human controls(30 cases).The level of IL-35 in serum was measured with enzyme-linked immunosorbent assay (ELISA).The subunit EBI3 and IL-12A of IL-35 mRNA expression in peripheral blood mononuclear cell(PBMC) were detected by SYBR Green real-time quantitative PCR (qPCR). Result：IL-35 level in AR group (251.22 ± 46.27pg/ml) was significant lower compared with normal control group(382.17 ± 25.41pg/ml),p0.01.The mRNA expression level of EBI3 in the patients with allergic rhinitis was significantly lower than that in the normal control group(p 0.001)，AR group EBI3 mRNA level was about half the normal control group.But the mRNA expression level of IL-12A in the patients with allergic rhinitis was not significant difference with normal control group(p0.05). Conclusion：The decrease of IL-35 and EBI3 mRNA in AR,indicated that IL-35 and EBI3 mRNA may play an important role in allergic rhinitis.
