禽偏肺病毒检测方法的建立及应用showedthattheRNAofaMPVdisplayedspecificpositivereaction,butothernucleotideextractedfromH9N2,NDV,IBV,E.coilandthetissuesofSPFwerenegative.Thesensitivitytestshowedthataslowas5pgofaMPV’sDNAcouldbedetectedbyDIG-labeledprobe.Thesamplesof605broilersand122duckswerecollectedfromflocksinShandongProvince,andthendotblothybridizationwasusedtodetectaMPV,theresultsshowedthatthedetectionratesofthetwokindsofbridswere36.59%and34.51%,respectively.SotheprobecouldbeusedinthedetectionofaMPV.ThisinvestigationalsoshowedthataMPVwaspresentinChinaandtheinfectionwascommoninbroilersandducksonsomefarmsinShandongProvince.3.EstablishmentandapplicationofIndirectELISAfordetectionofaMPVantibody.Thepurifiedandrecombinantproteinwasusedtocoatthe96poresELISAboardasantigen,whichwasusedtocoata96wellhighaffinityELISAplateasantigen.AseriestrialswereperformedforthegropedoptimalreactionconditionsoftheindirectELISAandtheresultswereobtainedasfollow:thebestcoatingquantityofantigenwas800ngperhole;Serumwasdiluted50times;Thebestcoatingconditionsofrecombinantproteinwas4℃overnightandthebestcoatingliquidwascarbonatebuffersolution(liquidpH9.6);Thebestblocksolutionwas5%skimmedmilkpowder,thebestblocktimewas1hat37℃;Thebestreactiontimeofserumwas90min;Thebestdilutedtimeofenzyme-linkedsecondaryantibodieswas1000forreaction60min;thebestreactiontimeofsubstratewas10min;thecut-offvalueofpositiveresultwas0.292(Cp=X+3SD),thenegativeresultwas0.258(Cn=X+2SD).Moreover,theestablishedindirectELISAmethodwiththeadvantagesofgoodrepeatabilityandspecificityofferedafoundationtoaMPVdiagnostickit.1139ChickenserumsfromsevenregionsofShandongweredetectedbythismethod.Theresultsshowedthatthegeneralpositiveratewas37.05%（422/1139）,includingthelowest11.41%(42/368)ofcommercialegg-chickenyerandthehighest62.95%(226/359)ofbroilerbreed.Thepositiverateofcommercialbroilerwas37.38%.TheresultsshowedthataMPVwaswidespreadinchickensofcertainpartsinShandongprovince.Keywords:Avianmetapneumovirus;Nucleicacidprobe;RT-PCR;IndirectELISA.4英文缩略表英文缩写bpDNARNA
