中南民族大学硕士学位论文 A b s t r a c t The optimal compatibility and the effect of concentration of the Chinese medicine ‘Longquan Compound’ on HepG-2 and EC-1 cell line were investigated. The cell proliferation curve and observation on the effect of the cells of HepG-2, EC-1 and EC-109 caused by the drugs were obtained through the method of MTT( methyl thiazolyl tetrazolium), and the optimal compatibility and concentration of the drugs were determined by fluorescence staining. It is shown that the seven-mixed drug(the major effective components abstracted from strychnine, mountain keats mushroom, common camptotheca fruit, solanum nigrum, stone cypress, natural indigo and tatarian aster root respectively) is effective between 500~1000 μ g/mL,but it can cause cells grow more between125~250μ g/mL,and the range for the seven-drug mixture(mixed before abstraction) and solanum nigrum are below 1000 μ g/mL for HepG-2 cell. While for EC-1,it is effective under the experimental concentration, the effective ranges of the three-mixed drug(the major effective components abstracted from strychnine,common camptotheca fruit and natural indigo respectively) are below 50 μ g/mL and between 150-400 μ g/mL; the seven-mixed drug’s ranges are below 25 μ g/mL and between 50-400 μ g/mL; for common camptotheca fruit, the effective ranges are below 25μ g/mL and between 50-200 μ g/mL. However, the concentrations of those three different drugs which can make EC-1 proliferating are between 50-100 μ g/mL，25-50 μ g/mL and 25-50 μ g/mL, respectively. While for EC-109,it is effective under the experimental concentration,the effective inhibitory concentration of solanum nigrum, mountain keats mushroom and strychnine are 15~100μ g/ml,25~50 μ g/mLand under 50μ g/ml.From the results of the more accurate method of fluorescence staining, it is found that the range for the seven-mixed drug is 5-10 μ g/mL; for the three-mixed drug, the range is 10-25 μ g/Ml,and for common camptotheca fruit, the rang
