Abstract In this research, the extract from Danshen (Traditional Chinese Medicine, TCM) was adopted to study its effects on inflammatory cells and tumor cells as well as their mechanisms. We focused on the comparison between complex active substances and single active substance of Danshen, aiming to provide a theoretical support for the development of Chinese Traditional Medicine. In order to determine the volume、concentration of ethanol and other factors which was used to extract the liposoluble constituent in Danshen, we used the orthogonal design to optimize the extract method. The concentration of the liposoluble constituent, under different conditions, was examined by HPLC. Finally, we obtained the satisfied method: using 8 volumes of 70% ethanol to extract the liposoluble constituent for 2 times and each time takes 2h. In the anti-inflammatory mechanism study, we utilized Mice RAW264.7 cell lines. Western Blotting and semi-RT-PCR were carried out to determine the expression of secretory Phospholipase A2 (sPLA2), intracellular Phospholipase A2 (cPLA2) and cyclooxygenase-2 (COX-2)，Aslo in the anti-tumor mechanism study, we chose the HepG2 cell lines for vitro experiment. The result shows that the extract form DanShen have a effect in the inhibition of sPLA2、 cPLA2、 COX-2 which expressed in inflammatory cells. And the apoptosis in tumor cells was detected by MTT after the treatment by different concentration of Danshen extract. There are so many pharmacological active substances in TCM. we are focusing on the different effection between the complex active substance (containing a variety of pharmacological active substance) and a single active substance (such as tanshinone II-A and cryptotanshinone) .This study aim to get some significant results that can give a support of the production machnism of TCM.
