NoDULATIoN NoDULATIoN AND MICROBIAL ECoLoGY OF SoYBEAN ENDORHIZOSHERE AFFECTED BY INoCULATED RHIZOBIAL HNOl AND MUl渔NT GXHNl00 ABSTRACT The Success of a rhizobial inoculant in the soil depends to a large extent on its capacity to compete against indigenous strains.With improving inoculant affection,ecological effection of indigenous microbial communities also need to be attended by rhizobium release to soil.Previous researches show:(1) endorhizosphere microbial communities vary in various plants;(2) endorhizosphere microbial communities vary in seasons;(3)some endorhizosphere microbial communities are not affected by both plant and season.With the wild type strain HN01 as the control,GXHNl00,a mutant(S.fredii.strain),Was used as soil inoculants for soybean in a field release experiment.The aim of this study is to investigate the differences of nodulation and indigenous microbial community in the root system soil after inoculating HN01 and GXHNl00. Inorder to gain a better understanding dynamics of bacterial communities of the endorhizosphere of the soybeaR,traditional culture—dependent,16S rDNA gene.based PCR followed by denaturing gradient gel electrophoresis(DGGE), and and 16Sr DNA library were used to evaluated the effects of these two swains influence the indigenous microbial community. Nodulation competition test showed that the nodulation occupancy of GXHNl00 WaS 9.4%while that ofthe HN01 was 49.4%.Data showed soybean plants increased the number of microbes.The DGGE paaerns suggested that the bacterial communities as determined from direct endorhizosphere DNA extracts were largely stable between inoculanted treatment and noninoculanted treatment. Phylogenetic tree of each 1 6Sr DNA library showed that the structl玳of endorhizosphere microbial community was very similar with each other.In our study,no inoculation却endent effects could be detected,while the soybean plants appeared to have a much stronger influence on the microbial community.
