Genetics Genetics Polymorphism of Sousa chinensis as Revealed by mtDNA Control Region，Cyt B and Microsatellite Markers Major：Biochemistry and molecular biology Name：Chert Hongshan Supervisor：Wu Yuping ABSTRACT For the understanding well on the characteristis of the population and the genetic diversity of Indo—Pacific humpback dolphin，Chinese white dolphin，Sousa chinens打 (Osbeck，1765)，the partial mitochondrial control—region and cytochrome B of the dolphin samples from Pearl River Estuary and Xiamen Waters were sequenced and the polymorphism of nine microsatellite loci of these samples was analyzed． In the D—loop，examination of a 730一bp segment revealed 34 variable sites defining 4 unique haplotypes，account for 4．66％of total nucleotide acid．Of these variable sites，32 were transition substitutions and 2 were transvertions，the ratio of transition／transvertion was 1 6：1，one insertion—deletion were detected also in the study．The average proportion of nucleotide A，T’C．and G were about 30|2％，313％，23．5％andl5．1％，respectively，the proportion of(A+T)(61．5％)was a lot more than that of(G+C)(38．5％)．There were total four haplotypes(A，B，C and D)defined，the most common haplotype(A)was shared by 69．23％of indo—pacific humpback dolphins sampled from the Pearl River Estuary and Xiamen waters∞=12)．The haplotypic diversity(h)and nucleotide diversity(Ⅱ)in Xiamen population were 0．667 and 0．0168．h andⅡin Pearl River Estury population were both 0．There was only one haplotype found in Pearl River Estuary population．yet four IV were were detected in those of the Xiamen waters(0=14．891)．The polymorphic sites of haplotype B were 29，accounting for 85．29％of the total numbers of polymorphic sites．A very low level of genetic variation in the population for mtDNA was indicated．The genetic variation of S chinensis inhabiting in the Xiamen waters was higher than that in the Pearl River Estuary(0 O)． The partial sequence 599 bp of the mitochondrial cytochrome B gene from 1 2 samples was examined．The
