II II 色进程中的作用,通过添加激活剂NaCl、抑制剂NaHSO3和酶催化产物葡萄 糖酸验证葡萄糖氧化酶的作用大小。实验结果显示,无论在有无外加刺激 物的条件下,葡萄糖氧化酶均与过氧化氢产率及脱色率的变化趋势一致, 说明葡萄糖氧化酶是A5p1产过氧化氢的酶系之一,也是主要的脱色酶之一; 但因为三者之间变化程度的不同,推测可能含有其他的产过氧化氢酶系作 用。而在马丁培养基中培养的A5p1未检测到葡萄糖氧化酶活力,初步推测 可能是因为GOD是由类黑精诱导产生的。根据类黑精机理探讨得出的结论, 探讨了糖蜜酒精废水中是否存在葡萄糖氧化酶及相应的脱色作用。结果发 现,糖蜜酒精废水脱色体系中也存在葡萄糖氧化酶的作用,酶活力变化趋 势也与过氧化氢产率及脱色率的一致。但是脱色率受葡萄糖氧化酶和过氧 化氢的影响较类黑精小,可能还存在其他酶系作用。 关键词:类黑精 黄曲霉 A5p1 H2O2 产率 葡萄糖氧化酶 生物脱色 III III DECOLORIZATION OF MELANOIDIN BY ASPERGILLUS FLAVUS A5P1 AND PRELIMINARY STUDY OF ITS MECHANISM ABSTRACT Molasses wastewater is a recalcitrant pollutant. At present, biological treatment is useful to reduce the color of molasses. Our group screened a strain, named Aspergillus flavus A5p1, has an ability to decolour the wastewater. In order to find out the mechanism of decolorization by A5p1, we chose melonoidin, a main pigment of molasses, as the object to study its mechanism, providing a theoretical basis for the bio-decolorization. At first, we determined conditions of culture of Aspergillus flavus A5P1, including the carbon source, concentration of carbon source, nitrogen source, concentration of nitrogen source and the initial pH. The results showed the rate of decolorization of melanoidin (concentration of 20%) by Aspergillus flavus A5P1 reached the highest value of 65% after 8 days of cultivation in the following conditions: 2.5%(w/v) of sucrose as the carbon source, 0.2%(w/v) of sodium nitrate as the nitrogen source and initial pH of 6.0. In the previous stage of the process of decolorization, the rates of decolorization of melanoidin and cells dry weight were positively related, and the pH was in the range of 5.0 to 6.0.
