·1790 · doi 10.3969/j.issn. 1673-4254.2014.12.17 J South Med Univ, 2014, 34(12): 1790-1793 临床研究 外周血及大肠癌组织外周血及大肠癌组织TGF-TGF-ββ11含量检测及临床意义含量检测及临床意义 黄永红，曹云飞，高 枫，张 森，张磊昌，龙军先 广西医科大学第一附属医院结直肠肛门外科，广西 南宁 530021 摘要：目的探讨转化生长因子（TGF-β1）在结直肠癌患者的表达情况及临床意义。方法 采用ELISA 方法检测50例结直肠癌患 者术前、术后，以及25 例健康志愿者的外周血TGF-β1表达水平差异，同时检测了结直肠癌肿瘤组织和癌旁正常大肠组织匀浆 上清的TGF-β1含量，分析TGF-β1浓度的变化与临床病理指标的关系。结果 结直肠癌患者术前血浆TGF-β1水平明显高于术 后组以及健康对照组，差异有统计学意义（P0.05）；肿瘤组织TGF-β1含量显著高于癌旁正常大肠组织（P0.05）；在CEA 阴性 及CEA 阳性患者中均可分别观察到类似的浓度改变。外周血TGF-β1水平与肿瘤大小呈正相关，随着肿瘤分期的进展，浓度明 显升高。结论 TGF-β1可联合CEA 检测，更好的为结直肠癌的临床诊断、评价预后和复发提供参考价值，同时为基于抗TGF-β1 的肿瘤免疫治疗提供参考依据。 关键词：TGF-β1；结直肠癌；CEA ；诊断；免疫治疗 Detection of transforming growth factor-β1 in colorectal cancer and its clinical significance HUANG Yonghong, CAO Yunfei, GAO Feng, ZHANG Sen, ZHANG Leichang, LONG Junxian Department of Coloproctological Surgery, First Affiliated Hospital of Guangxi Medical University, Nanning 530021, China Abstract: Objective To investigate the role of transforming growth factor β1 (TGF-β1) in patients with colorectal cancer. Methods Fresh peripheral blood were obtained from 50 patients (before surgery and at least one week after surgery) and 25 healthy donors in the morning. Fresh colorectal cancer tissues and the adjacent tissues (at least 5 cm from the tumor site) were obtained from patients undergoing tumor resection. The expression levels of TGF-β1 in the blood and tissue specimens were determined using ELISA. Results The plasma levels of TGF-β1 in patients with colorectal cancer were significantly higher than those in the healthy donors, and decreased after the surgery (P0.05). The tumor tissues expressed higher levels of TGF-β1 than the adjacent tissues from both CEA-negative and -positive patients.
