 Preparation, Characterization and Application of Lectin-Magnetic Particle Conjugates# YANG Ganglong, LI Zheng** 5 10 15 20 25 30 35 (Laboratory for Functional Glycomics, Life Science School, Northwest University, Xian 710069) Abstract: Magnetic particles have been developed for various applications in the biological and clinical determination. In this study, we synthesized the epoxy-activated magnetic particles to immobilization the lectins. The epoxy-activated magnetic particles were the superparamagnetic supports with a specific surface area of 96.29 m2/g. The each gram of the magnetic particles can be saturated with 186.31±5.16 mg Con A. The lectin-magnetic particle conjugates can isolate glycoproteins specially via the lectin’s carbohydrate recognition domain with well reproducibility. This method can be a good tool to be applied in the glycoproteomics research. Keywords: Biochemistrty and molecular biology; Expoxy-activated magnetic particles; Lectin- magnetic particle conjugates; Glycoprotein 0 Introduction With the development of microfluidic systems, magnetic particles have attracted considerable interest as a solid support. They are applied in a variety of biochemistry field, including immunoassays, support for enzymes, oligonucleotides, and bioaffinity sorbents[1]. Furthermore, the magnetic particles were widely used in the biomolecular separation, termed as magnetic separation technique. This technique is usually very simple, with only a few handling steps. All the steps of the purification procedure can take place in one single test tube or another vessel[2]. Lectins are frequently used in monitoring of glycosylation patterns of proteins or in the isolation of polysaccharides and glycoproteins. The magnetic particles are a good support for the lectin immobilization in the magnetic glycan separation technique[3].
