 PAGE \* MERGEFORMAT 15 HPLC fingerprint of TGP Research 【Abstract】 Objective To study the TGP HPLC fingerprint for scientific evaluation and effective control of its quality to provide a reliable method. Methods HPLC method, gradient elution, measured 21 samples of batch TGP. Chromatographic conditions: Hypersil C18 column (4.6 mm × 200 mm, 5 μm), flow rate 1.0 ml / min, column temperature 25 ℃, mobile phase A was acetonitrile; mobile phase B was 0.05% phosphoric acid in water, mobile phase A gradient elution (95% ~ 65% acetonitrile), analysis time was 60 min, time of 0,15,25,60 min, A (%) 95%, 86%, 86%, 65%. The results obtained, including 21 samples of batch TGP paeoniflorin (peak No. 7), including a total of 14 peaks. Cluster analysis and similarity analysis results are consistent. Conclusion TGP fingerprint characteristic and specific, can be used for full control over the quality of TGP to ensure that each batch of product uniformity. MethodsHPLC method was applied for quality assessment of Radix Paeoniae Alba. Column: Hypersil C18 column (4.6 mm × 200 mm, 5μm); flow-rate: 1.0 ml / min; column temperature: 25 ℃; mobile phase: acetonitrile (A) and H2O (acidified to 0.05% with phosphoric acid) (B ). The concentrations of solvent A were 95%, 86%, 86% and 65% at 0,15,25,60 min, respectively. ResultsThe HPLC chromatographic fingerprint of Radix Paeoniae Alba, showing 14 characteristic peaks, was established from 21 lots of Radix Paeoniae Alba products. The results of similarity analysis was the same as that of herarchical cluster analysis.ConclusionThe chromatographic fingerprint of Radix Paeoniae Alba with high specificity can be used to control its quality and assure to lot consistency.
