Methods: Indirect immunofluorescence assay were 86 cases of UC patients and 128 healthy persons serum ANCA testing, and polymerase chain reaction - restriction fragment length polymorphisms detected by oligonucleotide typing Shenzhen Han HLA-DPB1 gene polymorphism, using PCR technology for HLA-DPB1 gene exon 2 (exon 2) in vitro amplification; then use the Ban II, Fok I, Dde I, Rsa I, EcoN I, BstUI 6 kinds of restriction in enzyme digestion of amplified products, electrophoresis separation of fragments, according to the pattern of fragments to determine the corresponding genotype. Results: UC patients with ANCA positive rate was 53.5% in the control group were negative; UC patients with HLA-DPA1 allele frequency (0.49) and the control group (0.18), the difference was significant (P = 0.0032, RR = 5.681 , Pc = 0.033). Conclusion: HLA-DPA1 and the positive rate of ANCA in UC were significantly increased, HLA-DPA1 and ulcerative colitis have a certain association between ulcerative colitis patients and normal subjects there are differences in the immune genetic heterogeneity.
