Hypoglycemic mechanism of action of tea polysaccharide
[Abstract] Objective To study the mechanism of hypoglycemic effect of tea polysaccharides. Methods colorimetric determination of tea polysaccharides on α glucosidase (EC 3 2 1 20), and α amylase (EC 3 2 1 1) of the inhibitory activity was observed with rapid filtration of tea polysaccharide of rabbit intestinal brush-border vesicle glucose transport activity. The results show a strong tea polysaccharide α Glucosidase inhibitory activity, its α Glucosidase half-inhibitory concentration (IC50) value of 3 2g / L; tea polysaccharides on α amylase inhibitory activity was relatively weak, concentration of 1g / L, its right α amylase inhibitory rate of 10%; tea polysaccharides while significantly reducing the rabbit intestinal brush-border vesicle glucose transport capacity, the half-inhibitory concentration (IC50) value of 15g / L. Conclusion of tea polysaccharides inhibit glucosidase enzyme α α amylase and intestinal brush-border vesicle glucose transport capacity, which may delay the intestinal absorption of sugar.

Tea is a kind of polysaccharide extracted from the tea leaves to the multi-sugar compounds, with enhanced immunity, lowering blood pressure, lowering blood sugar, anti-radiation, anti-clotting, anti-thrombotic effects, etc. 〔 1,2〕 . China and Japan, the old folk have Paoyin rough treatment of diabetes, history of tea 〔 3〕 . Preliminary study confirmed that the polysaccharide is a rough old tea tea hypoglycemic main active ingredient 〔 4〕 , but the specific mechanism of action is unclear. In this paper, tea polysaccharide as the material to explore its α Glucosidase, α amylase and brush border vesicle glucose transport capacity of inhibition to reveal the hypoglycemic mechanism of tea polysaccharides.
