Followingproperactivation,na¨?ve“CD4lo”Tcellsdi?erentiateintoe?ectorTcellswithenhancedexpressionofCD4-“CD4hi” e?ectors.Autoimmunediabetes-proneNODmicedisplayauniquesetofantigen-experienced“CD4lo”Tcellsthatpersistafter primarystimulation.Here,wereportthatapopulationofsuchcellsremainedaftersecondaryandtertiaryTCRstimulationand producedcytokinesuponantigenicchallenge.However,whenNODblastswereinducedinthepresenceofrIL-15,thenumber of antigen-experienced “CD4lo” T cells was signi?cantly reduced. Clonal contraction, mediated in part by CD95-dependent activation-inducedcelldeath(AICD),normallyregulatestheaccumulationof“CD4hi”e?ectors.Interestingly,CD95expression wasdramaticallyreducedontheAICD-resistantNOD“CD4lo”Tcells.Thus,whileautoimmunediseasehasoftenbeenattributed totheengagementofrobustautoimmunity,wesuggestthattheinabilitytoe?ectivelycontracttheimmuneresponsedistinguishes benignautoimmunityfromprogressiveautoimmunediseasesthatarecharacterizedbychronicTcell-mediatedin?ammation. 1.Introduction necessarytopreventthelong-termaccumulationofactivated e?ectorTcells.Followingseveralroundsofclonalreplication and expansion, the antigen-speci?c T cell repertoire con- tracts to reestablish the homeostatic levels of lymphocytes [8].Thisclonalcontractionismediatedinpartbyactivation-
