This is particularly important in reconstitution work involving transfer of autoantibodies. Toaddressthisissue,wetransplantedhumanperipheralbloodmononuclearcells(PBMC)directlyinto thespleenofirradiatedNOD/SCIDmicedepletedofnaturalkillercellactivity.Within6weeks,na?¨ve t BcellsdifferentiatedintomemoryBcellsand,importantly,thenumbersofhumanCD138 plasma cells in spleen increased by100 fold after transplantation. Plasma cell numberscorrelated with the detectionofhumanIgMandIgGinserum,indicatingthathumanBcellshaddifferentiatedintomature plasmacellsinthemurinespleen.InadditiontoCD19 plasmacells,adistinctCD192plasmacell populationwasdetected,suggestingthatdownregulationofCD19associatedwithmaturationofplasma cellsoccurred.Whenpuri?edhumanBcellsweretransplanted,those?ndingswerenotobserved.Our results indicate that differentiation and maturation of human B cells and plasma cells can be investigatedbytransplantationofhumanPBMCintothespleenofNOD/SCIDmice.Themodelwillbe usefulforstudyingthedifferentiationofhumanBcellsandgenerationofplasmacells. t Keywords: Autoantibodies;Bcellontogeny;Reconstitution;NOD/SCIDmice INTRODUCTION differentiate into memory B cells and plasma
