Doppel(Dpl)isamembrane-boundglycoproteinmainlyexpressedinthetestisofadulthealthypeople.Itisgenerallyabsentinthe centralnervoussystem,butitscodinggenesequenceisectopicallyexpressedinastrocytomaspecimensandinderivedcelllines.In thispaper,weinvestigatedtheexpressionandthebiochemicalfeaturesofDplinapanelof49astrocytomaspecimensofdi?erent WHOmalignancygrades.Asaresult,Dplwasexpressedinthemajorityoftheinvestigatedspecimens(86%),alsoincludinglow gradesamples.Importantly,Dplexhibiteddi?erentcellularlocalizationsandalteredglycanmoietiescomposition,dependingon thetumorgrade.Mostlow-gradeastrocytomas(83%)showedamembrane-boundDpl,likehumanhealthytestistissue,whereas themajorityofhigh-gradeastrocytomas(75%)displayedacytosolicDpl.DeglycosylationstudieswithN-glycosidaseFand/or neuraminidasehighlighteddefectiveglycanmo
