Both elevation and reductioninintracellularcalciumcandisruptmitosisbymechanismsthatremainillde?ned.Inthisstudyweexploretheroleof transmembranevoltage-gatedcalciumchannels(CaVchannels)asregulatorsofmitosisinthemousecorticotrophcellline(AtT- 20).Wereportthatthenifedipine-sensitiveisoformCaV1.2islocalizedtothe“polewardside”ofkinetechoresduringmetaphase and at the midbody during cytokinesis. A second nifedipine-sensitive isoform,CaV1.3, ispresent at the mid-spindle zone in telophase,butisalsoseenatthemidbody.Nifedipinereducestherateofcellproliferation,and,utilizingtime-lapsemicroscopy, weshowthatthisisduetoablockattheprometaphasestageofthecellcycle.UsingFluo-4wedetectcalcium?uxesatsites correspondingtothemid-spindlezoneandthemidbodyregion.Anothercalciumdye,FuraPE3/AM,causesaninhibitionof mitosispriortoanaphasethatweattributetoachelationofintracellularcalcium.Ourresultsdemonstrateanovel,isoform- speci?clocalizationofCaV1channelsduringcelldivisionandsuggestapossibleroleforthesechannelsinthecalcium-dependent eventsunderlyingmitotic
