HindawiPublishingCorporation JournalofBiophysics Volume2008,ArticleID420963,5pages doi:10.1155/2008/420963 ResearchArticle CellVolumeandSodiumContentinRatKidneyCollectingDuct PrincipalCellsDuringHypotonicShock EvgenyI.Solenov InstituteofCytologyandGenetics,SiberianBranchofRussianAcademyofSciences,NovosibirskStateUniversity, Novosibirsk630090,Russia CorrespondenceshouldbeaddressedtoEvgenyI.Solenov,eugsol@bionet.nsc.ru Received3March2008;Accepted3July2008 RecommendedbyWalterSt¨uhmer The purpose of this study was to investigate the time course of the volume-regulatory response and intracellular sodium + concentration([Na ]i)intheprincipalcellsofratkidneyoutermedullacollectingduct(OMCD)epitheliaduringacuteswelling inhypotonicmedium.HypotonicshockwascreatedbyPBSdilutedwith50%ofwater.Changesincellvolumeweremeasured withcalceinquenchingmethod.Intracellularsodiumconcentrationwasstudiedwith?uorescencedyeSodiumGreen.Principal cellsofmicrodissectedOMCDfragmentsswelledveryfast.Thecharacteristictimeofswelling(τ1)was0.65± 0.05seconds,and thevolumeincreasedmorethan60%(92.9± 5.6and151.3± 9.8μm controlandpeakvolumescorrespondently,P.01).After 3 cellvolumereachedthepeakofswelling,theRVDbeganwithoutlagperiod.Thecharacteristictimeofvolumedecreasingto newsteady-statelevel(τ2)was8.9± 1.1seconds.Inhypoosmoticmedium,cellvolumestabilizedonhigherlevelincomparison withcontrol(110.3± 8.3μm ,P .01).Afterrestorationofthemediumosmolalitytonormotonic,cellvolumestabilizedon 3 + signi?cantlylowlevelincomparisonwithcontrollevel(71.4±6.1μm ,P.01).Duringthehypoosmoticshock,[Na ]idecreased 3 fromcontrollevelinisotonicPBStothelowlevelinhypoosmoticsolution(27.7±1.4and5.8±0.23mM,P.01).Calculationof sodiumcontentpercellhasshownthesigni?cantsodiumentryintothecells,whichcausedatemporaryincreasecorrelatedwith thepeakofcellvolumecausedbyswelling.Theconclusionismadethatinourmodelofhypoosmoticshock,swellingactivates transporterswithhighpermeabilityforNa thatprovidessodium?uxintothecells. +
