Severalwall macromoleculesof this alga are recognized by monoclonalantibodies speci?c forwallpolymer epitopes ofland plants.Immuno?uorescenceprotocolsusingtheseantibodiesmaybeemployedtolabelspeci?ccellwallconstituentsoflivecells. Fluorescentlabelingpersistsforseveraldays,andthisattributeallowsfortracingofwallepitopesinbothlong-andshort-term studiesofcelldevelopment.Quantitativeanalysisofsurfaceareacoveredbycellwallpolymersisalsoeasilyperformed.Weshow thatsigni?cantcellexpansioncausedbyincubationofcellsinlowlevelsofosmoticallyactiveagentslikemannitol,glucose,or sucroseresultsfromtheinabilityofcellstoundergocytokinesisbutdoesnotresultinsigni?cantchangestotheamountofnewcell wall.Wealsodemonstratethatcellscanbemaintainedforlongperiodsoftimeinculturemediumsupplementedwithspeci?ccell wall-degradingenzymeswherenotablechangestowallinfrastructureoccur.Theseresultsdemonstratethegreatpotentialvalueof Peniuminelucidatingfundamentaleventsduringcellwallsynthesisandmodulationinplantcells. 1.Introduction pectinsalongwithavarietyofproteoglycans/proteinsinclud- ingarabinogalactanproteins(AGPs),extensin,expansin,and multiple enzymes [5]
