EffectiveElicitationofHumanEffectorCD8+TCellsin HLA-B*51:01TransgenicHumanizedMiceafterInfection withHIV-1 YoshinoriSato,SayakaNagata,MasafumiTakiguchi* CenterforAIDSResearch,KumamotoUniversity,2-2-1Honjo,Kumamoto,Japan Abstract Humanized mice areexpectedtobe useful assmall animalmodels for in vivostudies onthepathogenesis ofinfectious diseases.However,itiswellknownthathumanCD8 Tcellscannotdifferentiateintoeffectorcellsinimmunodeficientmice + + transplantedwithonlyhumanCD34 hematopoieticstemcells(HSCs),becausehumanTcellsarenoteducatedbyHLAin themousethymus.WehereestablishedHLA-B*51:01transgenichumanizedmicebytransplantinghumanCD34 HSCsinto + + HLA-B*51:01transgenicNOD/SCID/Jak32/2mice(hNOK/B51Tgmice)andinvestigatedwhetherhumaneffectorCD8 Tcells would be elicited in the mice or in those infected with HIV-1 NL4-3. There were no differences in the frequency of late +/2 +/2 effectormemoryandeffectorsubsets(CD27lowCD282CD45RA CCR72andCD272CD282CD45RA CCR72 ,respectively) amonghumanCD8 TcellsandinthatofhumanCD8 TcellsexpressingCX3CR1and/orCXCR1betweenhNOK/B51Tgand hNOKmice.Incontrast,thefrequencyoflateeffectormemoryandeffectorCD8 Tcellsubsetsandofthoseexpressing + + + CX3CR1and/orCXCR1wassignificantlyhigherinHIV-1-infectedhNOK/B51Tgmicethaninuninfectedones,whereasthere wasnodifferenceinthatofthesesubsetsbetweenHIV-1-infectedanduninfectedhNOKmice.Theseresultssuggestthat hNOK/B51TgmicehadCD8 TcellsthatwerecapableofdifferentiatingintoeffectorTcellsafterviralantigenstimulation + + andhadagreaterabilitytoeliciteffectorCD8 TcellsthanhNOKones.
