NucleolarAccumulationofRNABindingProteins InducedbyActinomycinDIsFunctionalinTrypanosoma cruziandLeishmaniamexicanabutNotinT.brucei EzequielNa′zer,DanielO.Sa′nchez* InstitutodeInvestigacionesBiotecnolo′gicas-InstitutoTecnolo′gicoChascomu′s,UNSAM-CONICET,SanMart?′n,ProvinciadeBuenosAires,Argentina Abstract We have recently shown in T. cruzi that a group of RNA Binding Proteins (RBPs), involved in mRNA metabolism, are accumulatedintothenucleolusinresponsetoActinomycinD(ActD)treatment.Inthiswork,wehaveextendedouranalysis to other members of the trypanosomatid lineage. In agreement with our previous study, the mechanism seems to be conservedinL.mexicana,sincebothendogenousRBPsandatransgenicRBPwererelocalizedtothenucleolusinparasites exposedtoActD.Incontrast,inT.brucei,neitherendogenousRBPs(TbRRM1andTbPABP2)noratransgenicRBPfromT. cruziwereaccumulatedintothenucleolusundersuchtreatment.Interestingly,whenatransgenicTbRRM1wasexpressedin T.cruziandtheparasitesexposedtoActD,TbRRM1relocatedtothenucleolus,suggestingthatitcontainsthenecessary sequenceelementstobetargetedtothenucleolus.Together,bothexperimentsdemonstratethatthemechanismbehind nucleolarlocalizationofRBPs,whichispresentinT.cruziandL.mexicana,isnotfunctionalinT.brucei,suggestingthatithas beenlostorretaineddifferentiallyduringtheevolutionofthetrypanosomatidlineage.
