B?umeretal.FrontiersinZoology2012,9:15 /content/9/1/15 RESEARCH OpenAccess OpposingeffectsofNotch-signalingin maintainingtheproliferativestateoffolliclecells inthetelotrophicovaryofthebeetleTribolium DanielB?umer,NadiMStr?hleinandMichaelSchoppmeier* Abstract Introduction:EstablishmentofdistinctfolliclecellfatesattheearlystagesofDrosophilaoogenesisiscrucial forachievingpropermorphologyofindividualeggchambers.InDrosophilaoogenesis,Notch-signaling controlsproliferationanddifferentiationoffollicularcells,whicheventuallyresultsinthepolarizationofthe anterior-posterioraxisoftheoocyte.HereweanalyzedthefunctionsofTriboliumNotch-signalingfactorsduring telotrophicoogenesis,whichdiffersfundamentallyfromthepolytrophicovaryofDrosophila. Results:WefoundNotch-signalingtoberequiredformaintainingthemitoticcycleofsomaticfolliclecells. UponDeltaRNAi,folliclecellsenterendocycleprematurely,whichaffectsegg-chamberformationandpatterning. Interestingly,ourresultsindicatethatDeltaRNAiphenotypesarenotsolelyduetotheprematuretermination ofcellproliferation.Therefore,wemonitoredtheterminal/stalkcellprecursorlineagebymolecularmarkers. WeobservedthatuponDeltaRNAiterminalandstalkcellpopulationswereabsent,suggestingthatNotch-signaling isalsorequiredforthespecificationoffolliclecellpopulations,includingterminalandstalkprecursorcells. Conclusions:Wedemonstratethatwithrespecttomitoticcycle/endocycleswitchNotch-signalinginTribolium andDrosophilahasopposingeffects.WhileinDrosophilaaDelta-signalbringsaboutthefolliclecellstoleave mitosis,Notch-signalinginTriboliumisnecessarytoretaintelotrophicegg-chambersinan“immature”state. Inmostinstances,Notch-signalingisinvolvedinmaintainingundifferentiated(orpreventingspecialized)cellfates. Hence,theroleofNotchinTriboliummayreflecttheancestralfunctionofNotch-signalingininsectoogenesis. ThefunctionsofNotch-signalinginpatterningthefolliclecellepitheliumsuggestthatTriboliumoogenesis may-analogoustoDrosophila-involvethestepwisedeterminationofdifferentfolliclecellpopula
