Results:WeaimedtomanipulateoxygenstressperceptioninArabidopsisseedsbyoverexpressionofthenon- symbiotichemoglobinAtHb1underthecontroloftheseed-specificLeB4promoter.SeedsoftransgenicAtHb1 plantsdidnotaccumulateNOundertransienthypoxicstresstreatment,showedhigherrespiratoryactivityand energystatuscomparedtothewildtype.Globaltranscriptprofilingofseeds/siliquesfromwildtypeandtransgenic plantsundertransienthypoxicandstandardconditionsusingAffymetrixATH1chipsrevealedarearrangementof transcriptionalnetworksbyAtHb1overexpressionundernon-stressconditions,whichincludedtheinductionof transcriptsrelatedtoABAsynthesisandsignaling,receptor-likekinase-andMAPkinase-mediatedsignaling pathways,WRKYtranscriptionfactorsandROSmetabolism.OverexpressionofAtHb1shiftedseedmetabolismtoan energy-savingmodewiththemostprominentalterationsoccurringincellwallmetabolism.Incombinationwith metaboliteandphysiologicalmeasurements,thesedatademonstratethatAtHb1overexpressionimprovesoxidative stresstolerancecomparedtothewildtypewhereastrongtranscriptionalandmetabolicreconfigurationwas observedinthehypoxicresponse.
