InfluenceofSteroidogenicFactor-1GeneSilencingonRegulatingAldosteroneSecretionandCellProliferationinHumanAdrenalCorticocarcinomaH295RCellDepartmentofUrology,TongJiHospital,TongJiMedicalCollege,HuaZhongUniversityofScienceandTechnologyDoctoralCandidate:HuDongliangDoctoralAdvisor:Prof.ZhangXuAbstractINTRODUCTIONPrimaryaldosteronism(PA)isacommonendocrinedisordercharacterizedbyautonomoushypersecretionofaldosteroneandmulti-systemsymptoms.Thecausesofprimaryaldosteronismdeterminetheappropriatetreatmentsanddifferentresults.Aldosterone-producingadenoma(APA)andbilateralidiopathichyperaldosteronism(IHA)arethemostcommonsubtypesofPA.However,morethan90%ofAPAcanbecuredbyadrenalectomy,butIHAoftenneedsmedicaltreatment.AlthoughmanyauthorsmadegreatprogressinthepathologyandpreventionofPA,thedetailedpathogenesiswasstillnotveryclear.Steroidogenicfactor-1(SF-1)isanimportanttranscriptionfactor.AllsteroidogenicsynthaseshaveoneormoreSF-1bindingsequencesinthepromoterregion.SF-1participateswidelyinregulatingsteroidsynthesis,adrenalgrowth,tumorigenesis,andgonadaldevelopment.Theterminalstepinaldosteronebiosynthesisiscarriedoutbyaldosteronesynthase(CYP11B2),andangiotensin-II(AT-II)isthemostimportantregulatorofthem.Atpresent,therelatedstudiesontheroleofSF-1inaldosteronehormonesecretionandPApathogeniticmechanismweremerelyreportedandtheconclusionwasnotunified.Inrecentyears,peoplehaveconductedtheextensiveresearchintoCYP11B2andAPA,butmostofthemconcentratedincasereportsandindividualtargetdetectionintissues.However,therewereraredocumentsreportedincelllevel.SowestudiedtheroleofSF-1inthepathogenesisofPAthroughchangingitsgeneexpression.METHODSSixteenAPAsampleswerecollectedfrompatientswithclinicallyandpathologicallyconfirmedPA.Twelvenormaladrenalsamplescamefrompatientswho,duetorenalcarcinomaorrenalpelviccarcinoma,underwentadrenalectomyalongwithrenalectomy.SF-1expressionwasdeterminedbyWesternblottingandreal-timepolymerasechainreaction(PCR)method.ThreeDNAsequencesofSF-1whichcouldtranscriptesmallhairpinRNA(shRNA
