TIMP1对人牙周膜成纤维细胞增殖与ALP活性表达影响 　　[摘要] 目的:探讨金属蛋白酶组织抑制剂-1(TIMP-1)对人牙周膜成纤维细胞的增殖与碱性磷酸酶(ALP)活性表达的影响。方法:采用MTT和ALP活性检测法,观察不同浓度的TIMP-1在第24、48和72 h对体外培养的人牙周膜成纤维细胞(PDLF)的作用。结果:与对照组比较,1 ng/ml的TIMP-1在24、48和72 h,10 ng/ml的TIMP-1在48、72 h可显著促进PDLF的增殖;1 ng/ml的TIMP-1在24、48和72 h,10 ng/ml的TIMP-1在48 h对PDLC有显著增强ALP活性作用。结论:TIMP-1可促进人PDLF生长和分化。 　　[关键词] 牙周膜成纤维细胞;金属蛋白酶组织抑制剂-1;MTT;碱性磷酸酶 　　[中图分类号] R781.4 [文献标识码]A [文章编号]1673-7210(2010)01(c)-030-02 　　 　　Effects of TIMP-1 on human periodontal ligament fibroblasts proliferation and expression of ALP activity 　　GAO Xiaolin, LI Ying 　　(Department of Stomatology, the Second Affiliated Hospital of Shanxi Medical University, Taiyuan 030001, China) 　　[Abstract] Objective: To explore the biological effects of tissue inhibitor of metalloproteinase-1 (TIMP-1) on the proliferation and alkaline phosphatase (ALP) activity in human periodontal ligament fibroblasts (PDLF). Methods: Thiazoly blue (MTT) colorimetric assay and ALP activity analysis were used at different time points (24th, 48th and 72nd hours) to determine the proliferation and ALP activity of PDLF after different doses of TIMP-1 were added into the culture medium. Results: Compared with control group, 1 ng/ml TIMP-1 at 24th, 48th and 72nd hour, 10 ng/ml TIMP-1 at 48th and 72nd hour, significantly promoted the proliferation of PDLF. 1 ng/ml TIMP-1 at 24th, 48th and 72nd hour, 10 ng/ml TIMP-1 at 48th significantly increased the ALP activity of PDLF.
