慢性乙型肝炎患者血清IL12IL6水平变化和CD28表达研究 　　【摘要】 目的 探讨 IL-12、IL-6和淋巴细胞CD28分子在慢性乙型肝炎发病中的作机制。方法 严格按照病例入选标准选取 50例患者和 20例健康志愿者,每例空腹抽取外周静脉血 5 ml,3000 rpm离心10 min,吸取上层血清1.0 ml,放置-70℃ 冰冻保存备检,同时通过流式细胞仪检测所有患者外周血CD+28淋巴细胞所占比例。采用双抗体夹心ABC-ELISA法检测患者外周静脉血血清中细胞因子IL-12、IL-6的水平,比较慢性乙型肝炎患者与健康对照组及急性乙型肝炎之间Th1和Th2细胞因子表达的差异。结果 HBeAg(-)慢性乙型肝炎患者IL-12、IL-6水平显著高于健康对照组(P+28T淋巴细胞所占比例升高,显著高于健康对照组(P+28T淋巴细胞水平低于HBeAg(+)慢性乙型肝炎患者,差异有统计学意义(P28分子在乙型肝炎病毒的清除过程中起重要作用。 　　【关键词】 　　慢性乙型肝炎;白细胞介素-12;白细胞介素-6;CD28 　　 　　The study of the level of IL-12,IL-6 in serum and the expression of CD28 on lymphocyte in HBeAg(-)CHB patientsLI Lei,GAO Zhi-xing,JI Wan-sheng,et al.Department Internal Medicine of Weifang Medical College,Shandong Weifang 261042,China 　　【Abstract】 Objective To discuss the mechnism of T helper cells and their cytokines in patients with hepatitic B virus e antigene(HBeAg)negetive chronic hepatitis B(CHB).Methods We choosed forty patients and ten healthy volunteers strictly.We took 5 millilitres peripheral blood from each person and have three millilitres of them centrifuged for ten minutes at the speed of 3000rpm and took one millilitre serum to the refrigerator which the temperature is 70℃ below zero for future assay.We detected the production of IL-12 and IL-6 in all these specimens by avidin-biotin complex-enzyme linked immunosorbent assay(ABC-ELISA).Results The results of HBeAg negetive group showed that IL-12,IL-6 were all significantly upper adjusted(compared to the healthy control group:P+28T lymphocyte in HBeAg negetive CHB group was larger than that of the healthy control group,but it was smaller than that of HBeAg positive CHB group(P+28 T lymphocyte in the AHB group was largest in the four groups.Conclusion The disbalance of cell immunity and humoral immunity is existed in the HBeAg negetive CHB patients.The amounts of cytokins from Th1 cells have close relationship with the inflammatory activity of liver infected by HBV. 　　【Key words】 　　Chronic hepatitis B;HBeAg(-);Interleukin-12;Interle
