胰岛淀粉样多肽和P物质在不同发育阶段家兔直肠的表达及分布特点 洪 艳，梁文妹，韩 晶，胡 赟，夏白娟，李一欣 （550004贵阳，贵阳医学院基础医学院组织学与胚胎学教研室） [摘要] 目的 观察不同发育阶段家兔直肠内胰岛淀粉样多肽(islet amyloid polypeptide，IAPP)、P物质(substance P,SP)的表达及分布特点。方法 收集生后5、15、25、35、60 d及90 d家兔各10只。取直肠组织入Bouin液固定, 梯度酒精脱水,常规石蜡包埋, 制成连续切片。用免疫组织化学SABC法和图像分析法对IAPP、SP免疫反应阳性细胞的定位和表达进行观察。结果 生后5 d家兔直肠内可见少量散在分布IAPP阳性细胞，多位于上皮；35 d IAPP阳性细胞的数量呈明显上升趋势，至60 d达到最多(P0.05)，90 d时减少(P0.05)，主要分布于固有层。图像分析测得IAPP阳性细胞的平均灰度值随家兔的生长发育逐渐降低，60 d达最低(P0.05)。SP阳性细胞于生后15 d始出现，35 d后细胞数量明显增多(P0.05)，60 d达最高(P0.05)。其平均灰度值也于35 d开始下降(P0.05)，于60 d达最低水平(P0.05)。结论 IAPP、SP在直肠的表达随家兔的生长发育而逐渐变化，推测两者参与调节生后发育期间家兔直肠消化功能的成熟过程。 [关键词] 胰岛淀粉样多肽；P物质；直肠；生后发育；家兔 [中图法分类号] [文献标志码] A Expression profiles of islet amyloid polypeptide and substance P in rabbit rectum of rabbits during the postnatal development Hong Yan，, Liang Wenmei?，, Han Jing, ，Hu Yun, Xia bBaijuan，, Li Yixin（ (Department of Histology and Embryology, School of Basic Medical Sciences, Guiyang Medical College, Guiyang, Guizhou Province, 550004, China） [Abstract] Objective To investigate determine the expression profiles and distribution features of islet amyloid polypeptide (IAPP) and ,substance P （(SP）) in the rectum of rabbits during the postnatal development. Methods Sixty rabbits were equally divided into 6 groups according to their age, of the 5th-, 15th-, 25th-, 35th-, 60th-, and 90th- days groups. Their rectum tissue were collected, fixed in Bouin solution, embedded in routine paraffin and then made into serial sections. The expressions of IAPP and SP were observed by immunohistochemical SABC staining and image analysis . Results In the 5 5th-days group, a few IAPP positive-cells were scattered into the mucosa epithelium of rectum .
