PAGE PAGE III Abstract Objective: To study the effect of mifepristone combined with chemotherapy drug cisplatin and 5-fluorouracil on human cervical cancer cells in vitro. To find a new combination to inhibit cancer cell growth by inducing apoptosis and investigate its molecular mechanicsm. Methods: human cervical cancer HeLa、SiHa cell lines were cultured in vitro. Cells were treated with various concentration of MIF or chemotherapy drug or both. Cell proliferation of HeLa、SiHa were evaluated by MTT assay; The apoptosis rate and cell cycle of cells were determined by PI staining flow cytometry; The levels and their changes of HPV E6、P53、survivin 、Bax and Bcl-2 proteins of cells were investigated by Western blot. Results: (1)MTT assay indicated that MIF could inhibit the proliferation of SiHa cells in a concentration dependent and time dependent positive and which action time excessd 48h could inhibit the proliferation of Hela cells, while the proliferation of Hela cells was not affected by low concentrations(0.625,1.25,2.5,5,10 μg / mL) within 24h compared with the control group(P 0.05); Cells proliferation was inhibited (P 0.05) with different concentrations of DDP ,5-Fu in a dose-and-time dependent manner after evaluating the vitality of human cervical cancer by MTT assay; DDP ,5-Fu combined with low concentrations of MIF enhanced the proliferation inhibition rate of HeLa、SiHa cells (P 0.05). (2) PI staining flow cytometry data showed the apotosis rate of combination therapy significantly increased compared with single drug (P 0.05), but cell cycle of HeLa and SiHa cells did not be changed regularly. (3) Western blotting demonstrated that the expression of P53 protein increased in all combined groups, and the expression of HPV E6 protein decreased in HeLa cells treated with MIF combined with DDP/5-Fu; However ,the expression of survivn protein were different between MIF combined with different chemotherapy drug .
